Abstract : Introduction: Acute lymphoblastic leukemia (ALL) is a blood disease caused by stem cell malignancy due to blocked lymphoblast variation. Researches stated that ALL is frequently found among children and it most likely the main cause of death from cancer prior the age of 20. Interestingly, several genetic and epigenetic markers have been identified which can be used to predict patient prognosis and treatment response. DNMT3A and IDH1/2 genes are responsible for DNA methylation process by relocating a methyl group to the fifth carbon in cytosine guanosine (CpG) dinucleotide. Mutation of these genes can cause aberrant methylation of the genome CpG islands (CGIs), which leads to increase of suppressed oncogenes transcription or repression of active tumor suppressor gene transcription. Materials and Methods: Here, we performed a direct sequencing to investigate common mutations of DNA methylation-associated genes, DNMT3A (R882), IDH1 (R132) or IDH2 (R172 and R140) in 30 childhood ALL patients. Results: The findings indicated that there were no DNMT3A, IDH1 or IDH2 mutations in these patients. Conclusion and Recommendations: In this study, we aimed to detect the incidence of DNMT3A, IDH1 and IDH2 mutations occurring in children suffering from ALL, and determine their usefulness as prognostic indicators. In summary, previous studies that have been conducted in different regions or countries have led to different results. Therefore, since Saudi Arabia is a multiethnic society, we cannot generalize these findings.
